Analysis of 5' flanking sequences and intron-exon boundaries of the rat prolactin gene.

چکیده
A rat genomic DNA clone containing the 5’ flanking region, three exons, two introns, and a portion of a third intron of the rat prolactin gene was isolated and characterized. Sequence determinations were used to identify the exon-intron boundaries and analyze the DNA region upstream from the initiator methionine codon. Another genomic DNA clone that we previously characterized (Gubbins, E. J., Maurer, R. A., Lagrimini, M., Erwin, C. R., and Donelson, J. E. (1980) J. Biol. Chem. 255, 8655-8662) contains two additional exons and the 3’ flanking region of the prolactin gene. These five exons account for the complete coding sequence of rat preprolactin. Their relative locations in the two genomic DNA clones indicate that the five exons and four intervening sequences (introns) of the rat prolactin gene comprise approximately 10 kilobases of DNA. The sequence at the Intron A-Exon I1 boundary was found to contain two separate splicing sites offset by three nucleotides. These two splicing sites can explain a discrepancy of one codon in two published preprolactin cDNA sequences. The 5’ terminus of prolactin mRNA was mapped using the S1 nuclease protection procedure. The results suggest that transcription is initiated 54 nucleotides upstream from the initiator methionine codon. Analysis of the sequence preceding this site reveals (i) the sequence TATAAA at positions -21 to -22 which, by analogy with other systems, is likely involved in initiation of prolactin gene transcription and (ii) the palindrome, TGATTATATATATATTCA, at positions -62 to -45 which may be a site involved in the regulation of prolactin gene transcription.

متن کاملGenetic Variations in Exon 3 of VWF Gene in Patients with Von Willebrand Disease (VWD) from South-West Iran
Abstract Background Von Willebrand disease (VWD) is an autosomally inherited bleeding disorder with the prevalence of 1% based on population studies. The disease phenotype is due to quantitative and structural/functional defects in Von Willebrand Factor (VWF) which is a glycoprotein with essential role as a carrier of FVIII in circulation and also it serves the function as hemostasis regulato...
متن کاملIdentification of Novel Mutations in IL-2 Gene in Khorasan Native Fowls
The intron-exon structure of Khorasan native fowl interleukin-2 (IL-2) was investigated. For this purpose, twenty chickens were selected from the Native Fowl Breeding Station of Khorasan province, and genomic DNA was extracted using a modified conventional DNA extraction protocol. An 875 bp fragment of IL-2 was successfully amplified, including a small part of the promoter, exon 1, intron 1, an...
متن کاملFine structure and evolution of the rat serum albumin gene.
The exons, their boundaries, and approximately half of the intronic deoxyribonucleic acid of the rat serum albumin gene were sequenced. In addition to the 14 exons identified earlier by R-loop analysis, a small exon was detected between the "leader" exon (Z) and exon B. The leader exon encoded the 5'-untranslated portion of albumin messenger ribonucleic acid and the "pre-pro" oligopeptide prese...
متن کاملStructural organization of glycophorin A and B genes: glycophorin B gene evolved by homologous recombination at Alu repeat sequences.
Glycophorins A (GPA) and B (GPB) are two major sialoglycoproteins of the human erythrocyte membrane. Here we present a comparison of the genomic structures of GPA and GPB developed by analyzing DNA clones isolated from a K562 genomic library. Nucleotide sequences of exon-intron junctions and 5' and 3' flanking sequences revealed that the GPA and GPB genes consist of 7 and 5 exons, respectively,...
